What We Do Not Know About Cells

The intracellular reactions of E.coli chemotaxis constitute one of the bestunderstood behavioral responses. We know the principal proteins responsible, their structures and biochemical properties and how they enable the bacterial cell detect and respond to attractants and repellents[1]. Numerous computer simulations have been developed that reproduce essential features of the response[2, 3]. In a sense, therefore, bacterial chemotaxis serves as a benchmark of how well we understand living cells. Unfortunately, I will argue, close examination of this topic reveals that we are still a long way from the finishing line. Quite apart from specific issues, such as how the flagellar motors are driven by a flux of protons or how clusters of receptors in the cell membrane parse incoming information, there are also more general limitations. There is a level of molecular detail that we are presently unable to access for any cellular process, and this is especially true for higher organisms.
